摘  要:目的采用HPLC法测定拉米夫定中的右旋体。方法采用CyelobondI2000色谱柱(250mm×4.6mm,5μm),甲醇-0.1mol-L-醋酸铵(5:95)为流动相,流速0.8ml·min-1,柱温10℃~30℃,检测波长270nm。结果拉米夫定的光学异构体得到了基线分离,其右旋体0.5~5μg·ml。与峰面积的线性关系良好(r=0.9995),最低检测限为1.5ng。结论所建方法准确、简便、快速,适合于拉米夫定中右旋体的测定。OBJECTIVE To develop an HPLC method for determination of dextroisomer in Lamivudine. METHODS A cyclobond I 2000 column (250 mmx 4. 6 mm ,5 μm) was used with the mobile phase of methanol -0. 1 mol· L- 1 ammonium acetate (5 : 95 ) at a flow rate of 0. 8 ml-min-1 and the temperature of the column was 10℃ -30℃ with a detective wavelength at 270 nm. RESULTS The separation of both enantiomers of Lamivudine could be achieved baseline resolution. The calibration curve for Lamivudine dextroi- somer exhibited an excellent linear relationship with a correlation coefficient of 0. 9995 in concentration range of 0. 5 - 5.0 μg· ml -1 The detection limit was 1.5 ng for lamivudine dextroisomer. CONCLUSION The method is accurate, simple, quick and suitable for determination of Lamivudine dextroisomer.
