膀胱移行细胞癌中候选肿瘤干细胞标记物USP22mRNA水平定量分析及其与肿瘤分级的关系

摘　　要：目的:研究肿瘤干细胞标记物USP22 mRNA在不同级别膀胱移行细胞癌和正常膀胱黏膜中的表达差异,并探讨其与肿瘤分级,临床分期的关系。方法:分别抽提不同级别的膀胱移行细胞癌组织标本和正常膀胱黏膜组织标本的总RNA,经RT为cDNA后,采用qRT-PCR法检测USP22 mRNA的表达情况。结果:正常膀胱黏膜组织中USP22的表达明显低于膀胱肿瘤组织,几乎不表达;不同级别膀胱肿瘤组织中,低度恶性潜能的乳头状尿路上皮癌和低级尿路上皮癌中USP22的表达要明显高于其在高级尿路上皮癌中的表达;特别是Ⅲ级膀胱移行细胞癌组,USP22的表达仅略高于其在正常膀胱黏膜中的表达。结论:USP22可能是正常干细胞向肿瘤干细胞恶变的关键调节分子,可能成为一个新的诊断移行上皮癌侵袭性的有效指标。干扰其蛋白质合成将成为一个抗肿瘤生长的新靶点。Objective：To investigate the expression of the putative Cancer Stem Cell Marker USP22 mRNA in different grades of transitional cell carcinoma of bladder （TCCB） and the normal bladder, and to discuss the relationship between USP22, and the grade of tumor or clinical stage. Methods：The total RNA was extracted from the different grades of TCCB and the normal bladder tissue respectively, and the mRNAs were reversely transcribed into cDNA by reverse transcription PCR. The expression levels of USP22 mRNA were detected according to the standard curve using real-time fluorescence quantitative PCR. Results：The expression of USP22 in normal bladder was markedly lower than that of the bladder tumor. Among the bladder tumors, the expressions in the low malignant potential papillary urothelial neoplasm and in the low grade urothelial carcinoma were much higher than that of the high grade urothelial carcinoma. Especially, for the TCCB grade 3, the expression was only slightly higher than that of the normal bladder. Conclusions：The gene of USP22 maybe play an important role in the progression of stem ceils transforming into cancer stem cells, and may become a new marker in the diagnosis of TCCB. Inter feting the synthesis of the protein which is encoded by USP22 would become a novel anti tumor target .
