摘  要:目的:对食管贲门双源癌和单发食管癌及单发贲门癌患者血清蛋白质组和癌组织基因变化差异进行分析,筛选食管癌、贲门癌关键候选蛋白和基因。方法:采用表面增强激光解析离子化-飞行时间-质谱(SELDI-TOF-MS)与比较基因组杂交(CGH)技术分析比较食管贲门双源癌(4例)和单发食管癌(107例)、单发贲门癌(86例)患者血清蛋白质组和癌组织全基因组。结果:根据SELDI-TOF-MS蛋白质组所筛选的候选蛋白与CGH所筛选的基因进行一致性对比分析发现:双源癌中食管癌血清蛋白质组和癌组织CGH变异基因位点相一致的蛋白有14种(52%,11/21);双源癌中贲门癌与单发贲门癌位点相一致的蛋白有2种(Beta-defensin 123,GARP);双源癌、单发贲门癌和单发食管癌3者相一致的蛋白有1种(sPLA2-ⅡA)。结论:所发现的基因变异位点与蛋白质组候选蛋白对比相一致的17种蛋白可能是食管和贲门癌变关键候选蛋白和基因。Aim: To identify the key candidate genes and proteins related with esophageal and gastric cardia carcinogenesis by differential analysis on the patients with single esophageal or gastric cardia cancers or both from the same patient and proteomics. Methods: The surface-enhanced laser description/ionization time-of-flight mass spectrometry (SELDI- TOF-MS) and comparative genomic hybridization (CGH) were applied to the 4 patients with concurrent esophageal and gastric cardia cancers (CC), 107 patsents with single esophageal cancer(EC) and 86 patients with gastric cardia cancer. Results : Based on the consistent analysis by CGH and SELDI-TOF-MS, 14 cross proteins were identified both by CGH and SELDI-TOF-MS on EC from CC and single EC patients (52% , 11/21 ) ; 2 cross proteins (Beta-defens in 123, Gamma- aminobutyric acid receptor-associated protein) on gastric cardia adenocarcinoma (GCA) from CC and single GCA patients, and 1 cross protein (Pho-spholipase A2, membrane associated) from the patients with CC, single EC and single GCA were identified. Conclusion: The seventeen cross proteins identified in this study may be a group of key candidates related with esophageal and gastric cardia carcinogenesis.
