摘 要:目的:分析宫颈鳞癌HeLa229细胞中核转录因子(NF-κB)的激活状态及其与抗凋亡基因Bcl-2表达的相关性。方法:采用免疫细胞化学法和Western Blot法检测宫颈鳞癌HeLa229细胞中NF-κB亚单位p50和p65及其下游调控基因Bcl-2的蛋白表达,RT-PCR法检测宫颈鳞癌HeLa229细胞中p50和p65 mRNA的表达。EMSA法检测HeLa229细胞核中NF-κB与DNA的结合活性。结果:宫颈鳞癌HeLa229细胞质中NF-κB的亚单位p50和p65均有表达,p50/p65在细胞核中具有较高的DNA结合活性。RT-PCR结果表明,p50和p65的mRNA在细胞质中也存在表达,并且在HeLa229细胞中也检测到Bcl-2蛋白的表达。结论:在宫颈鳞癌HeLa229细胞中存在有NF-κB及其下游基因Bcl-2的表达,提示激活的NF-κB信号通路可能在宫颈鳞癌发生发展及抗凋亡中起重要作用。Aim :To analyze the relationship between activated NF-κB and the expression of Bcl-2 in cervical carcinoma HeLa229 cell line. Methods:Immunoeytochemistry and Western Blot were used to determine the protein levels of the subunits of NF-κB, pSO and p65, Bcl-2 in cervical carcinoma HeLa229 cell line. RT-PCR was used to detect the mRNA level about pS0 and p65. Nuclear proteins from the HeLa229 cell line was extracted to evaluate the DNA-binding activity by electrophoretic mobility gel shift assay (EMSA). Results: Cervical carcinoma HeLa229 cell line constitutively expressed pSO and p65 both in protein level and mRNA level. In addition, the results of EMSA analysis for nuclear protein extract showed high DNA-binding activity of p50 and p65. The protein level of Bel-2 was also detected in cervical carcinoma He- La229 cell line. Conclusion: In cervical carcinoma HeLa229 cell line, there is the expression of NF-κB and its downstream gene Bcl-2, suggesting that NF-κB may play a critical role in carcinogenesis and anti-apoptosis of the cervical carcinoma.
