摘 要:目的建立蛋白质芯片技术检测先天性巨结肠患儿血清蛋白质,筛选特异的蛋白质标记物,构建用于先天性巨结肠患儿早期筛选及普查的血清蛋白质指纹图谱模型。方法应用蛋白质指纹图谱分析仪(表面增强激光解析-电离-飞行时间质谱仪,SELDI-TOF-MS),测定62例血清标本(先天性巨结肠患儿42例,健康儿童20例),并结合生物信息学方法支持向量机分析数据。结果筛选出3个m/z位于3221.7、5639.2、6884.2的蛋白质标记物构建用于先天性巨结肠早期筛选及诊断的血清蛋白质指纹图谱模型。经留一法交叉验证,区分先天性巨结肠和正常小儿的血清蛋白质指纹图谱模型的特异性为100%,敏感性为100%。结论应用SELDI-TOF-MS结合生物信息学方法支持向量机构建的血清蛋白质指纹图谱模型在早期筛选及普查先天性巨结肠患儿的诊断中较以往的传统方法具有较高的敏感性和特异性,值得进一步研究与应用。[Objective] To set up a method for the detection of the serum protein by using the protein chip technology for exploration of serum protein finger print pattern models in early screening and diagnosis of Hirsehsprung's disease (HD). [Methods] The protein mass spectrograms was detected by the surface-enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS) technology combined with bio-informatics methods (support vector machine SVM) in 62 serum samples (42 from HD patients, and 20 from healthy individuals). [Results] Three protein markers whose m/z positioned at 3221.7, 5639.2, and 6884.2 were detected from the fingerprint ehromatogram model of serum protein for early screening and diagnosis of Hirschsprung's disease. The markers had 100% sensitivity and specificity. [Conclusion] In comparison with the traditional methods, the fingerprint chromatogram model of serum protein using SELDI-TOF-MS combining SVM has higher sensitivity and specificity in early screening and diagnosis of HD and should be further studied.
