摘 要:目的建立能够模拟肿瘤生长、侵袭转移动态全过程的动物模型,利用该模型探讨骨桥蛋白在肿瘤生长转移过程中的作用。方法Lewis肺癌(LLC)在C57BL/6小鼠体内连续传代,观察第2代(早期组)、第8代(中期组)和第15代(晚期组)荷瘤小鼠肿瘤生长和肺转移的情况,采用免疫组化法检测各组荷瘤鼠肿瘤组织中骨桥蛋白(OPN)和微血管密度(MVD)的表达。结果①随传代次数的增加肿瘤生长速度加快、侵袭力增强,肺转移率升高。②OPN和MVD在早、中、晚三组小鼠瘤组织中的表达均呈升高趋势(P〈0.05),且二者的表达呈正相关。结论OPN可能通过促进肿瘤血管生成的途径加速肿瘤的生长转移。Objective To establish an animal model that can simulate the dynamic process of tumor progression and metastasis and to explore the important role of osteopntin in this process. Methods LLC was maintained by serial passage in C57BL/6 mice, to observe the dynamic process of tumor progression and metastasis in the second generation, the eighth generation and the fifteenth generation, immunohistochemistry was used to detect the expression of OPN and MVD in tumor in each group. Resluts (1)With the numbers of passage increasing, the tumor' s growth velocity speeds up, the invasive power was gradually enhanced and the rate of pulmonary metastasis steps up. (2)The expression of OPN and MVD were different in each group. There was statistically significant correlation between the expression of OPN and MVD. Conclusions It indicates that OPN might participate in angiogenesis of tumor and promote the growth and metastasis of tumor.
