广西壮族食管癌患者FHIT基因表达及与Bcl-2表达的相关性研究

摘　　要：目的探讨FHIT基因在广西壮族食管癌发生、发展中的作用及其与Bcl-2表达的关系。方法应用免疫组化SP法检测FHIT基因和Bcl-2在广西壮族10例正常食管黏膜组织、80食管癌组织中的表达。结果FHIT基因在食管癌组织中的阳性表达显著低于正常食管黏膜组织(P<0.01);FHIT基因低表达与食管癌组织学类型无关(P>0.05),而与分化程度、浸润程度、有无淋巴结转移有关(P<0.05);食管癌中FHIT基因表达与Bcl-2蛋白表达有显著相关性(P<0.05)。结论FHIT基因表达缺失在广西壮族食管癌中是频发事件,FHIT基因参与食管癌的发生发展,是一个候选抑癌基因。在广西壮族食管癌患者FHIT基因的表达与Bcl-2的表达密切相关,FHIT基因和Bcl-2广西壮族食管癌发生、发展中起协调作用。Objective To study the expression of FHIT and it＇s relationship with expression of Bcl-2 in esophageal carcinoma in the Zhuang nationality in Guangxi. Methods The expression of FHIT and Bcl-2 studied by immunohistochemical technique （SP method） in 10 cases of normal esophageal mucosa and 80 cases of esophageal carcinoma. Results The FHIT expression rate of esophageal carcinoma was significant lower than that of normal esophageal mucosa （ P 〈 0. 01 ）. In loss or reduction expression of FHIT gone to histological type no correlation was found （P 〉 0.05 ）, but FHIT gene correlated with degree of the differentiation, invasion of depth tumors and lymph node metastasis （P 〈 0.05 ）. The expression of FHIT was associated with Bcl-2 expression in esophageal carcinoma （ P 〈 0.05 ）. Conclusion Loss or reduction of FHIT protein expression is a frequent event and FHIT gene involves in carcinogenesis of esophageal carcinoma and it may be a tumors suppressor gone candidate in esophageal carcinoma;FHIT expression was strongly associated with Bcl-2 expression in esophageal carcinoma. FHIT and Bcl-2 maybe play an coordinated role in tumor genesis and development of esophageal carcinoma.
