摘 要:目的:探索NO与偏头痛中枢敏化的关系。方法:体重为2~4kg的短毛猫12只,随机分为反复刺激组(n=4)、急性刺激组(n=4)、假手术对照组(n=4)三个组。应用免疫组化技术,行延髓和上颈髓冰冻切片nNOS染色。结果:nNOS免疫反应阳性神经元主要位于延髓三叉神经脊束核尾侧部浅层和C1、C2后角的I、Ⅱ层。假手术对照组动物免疫反应阳性神经元较少;急性刺激组和反复刺激组免疫反应阳性神经元较假手术对照组明显增多;急性刺激组和反复刺激组免疫反应阳性神经元数无显著差异。结论:NO在偏头痛的痛觉传递中起重要作用,可能是偏头痛中枢敏化产生和维持的重要因素。Objective:To explore the role of NO on the central sensitization of migraine. Methods:12 cats weighted 2.0 - 4.0 kg were randomly divided into three groups : sham-operation, acute stimulation and iterative stimulation groups. The brainstem and the superior cervical spinal cord were blocked and cut in coronal sections on a freezing microtome. The expression of nNOS immuno-reactivities was detected with immunohistochemical methods. Results:The nNOS immunoreactive positive neurons were observed in laminae Ⅰ and Ⅱ of the trigeminal nucleus caudalis and the dorsal horn of the C1 and C2 segment of the cervical cord. The number of nNOS immunoreactive positive neurons increased markedly in the stimulation groups comparing with the corresponding sham-operation group. There was no significant difference on the number of nNOS immunoreactive neurons between acute stimulated group and iterative stimulated group. Conclusion : NO plays an important role in the pain tansmission of migraine. It perhaps plays an important role in initiation and maintenance of the central sensitization in migraine.
