摘 要:哺乳动物的胃泌素释放肽(gastrin-releasing peptide receptor,GRP)在许多肿瘤中是一个自分泌生长因子,他涉及许多人类恶性肿瘤的发病机制和进展,在许多人类肿瘤中他的受体(GRPR)也是高表达的,这使得GRPR成为一个实验和临床治疗的新分子靶点.这里GRPRs可以作为细胞毒素、免疫毒素或者有放射活性化合物的有效传递体.而且,通过反义寡核苷酸、RNA干扰和GRPR拮抗物来封闭胃泌素释放肽信号通路后,结果都呈现出满意的抗肿瘤活性.The mammalian gastrin-releasing peptide (GRP), known as autocrine growth factors in tumors, is involved in the pathogenesis and progression of many human malignant tumors, and high expression of its receptor, GRPR, in a large spectrum of human cancers gives support to the conclusion that GRPR is a new molecular target in experimental and clinical cancer therapy. GRPRs may be potential carriers for cytotoxins, immunotoxins or radioactive compounds. Moreover, blocking gastrin-releasing peptide receptor signaling pathways by means of antisence oligonucleotide, RNA interference and its antagonists has exhibited impressive anfitumor activity.
