黄芩甙对重症急性胰腺炎大鼠胰腺ICAM-1和P-选择素表达的影响

摘　　要：目的观察黄芩甙对重症急性胰腺炎(SAP)大鼠胰腺组织ICAM-1和P-选择素表达的影响情况。方法将45只大鼠随机分为假手术组、模型组、黄芩甙组,每组15只。假手术组进腹后仅翻动胰腺和十二指肠后关腹,模型组、黄芩甙组在造模成功后分别给予生理盐水和黄芩甙。术后3,6,12 h,观察大鼠胰腺的病理变化,测定胰腺组织中髓过氧化物(MPO)和肿瘤坏死因子-α(TNF-α)的含量,用免疫组化方法检测胰腺组织ICAM-1和P-选择素的表达情况。结果术后3,6,12 h,模型组和黄芩甙组的病理评分均显著高于假手术组(P均<0.05),且模型组和黄芩甙组6,12 h的病理评分均显著高于3 h(P均<0.05),但2组评分在6 h和12 h间无显著性差异;术后3,6,12 h,模型组胰腺组织中MPO和TNF-α较假手术组显著降低(P均<0.05),黄芩甙组胰腺组织中MPO和TNF-α较模型组显著降低(P均<0.05);术后各时间点,模型组ICAM-1和P-选择素随时间延长显著增多,黄芩甙组则随时间延长而显著减少。结论黄芩甙可能通过抑制中性粒细胞的浸润,减轻ICAM-1和P-选择素的表达而发挥对胰腺组织的保护作用。Objective It is to observe the effect of baiealin on the expression of ICAM- 1 and P - selectin in pancreatic tissue in rats with severe acute pancreatitis（SAP）. Methods 45 rats were randomly divided into sham operation group （group SO）, model group （group M） and baicalin-treated group （group B）, and each group had 15 rats. Rats in group SO were suffered from opening and suturing abdomen with fliping heart sweetbread and duodenum, and were given no drugs. Rats in the other two groups were given baicalin and normal saline respectively after model successfully established. At 3, 6 and 12 hours after operation, the pathological changes of pancreas were observed, the content of MPO and TNF- α in pancreas were determined, and immunohistochemieal examination of ICAM - 1 and P - selectin were performed. Results Compared with group SO, pancreatic histological scores of rats in group B and M increased significantly at 3, 6 and 12 hours after operation （ P 〈 0.05）, and pancreatic histological scores at 6 and 12 hours were higher than those at 3 hours after operation in group B and M （P 〈 0.05）, but there was no significant difference in histological scores between 6 and 12 hours in group B and M （P〉0.05）. At 3, 6 and 12 hours after operation, the content of MPO and TNF- α in rat pancreatic tissue of group B decreased significantly compared with that of group M （ P〈 0.05）. The expression of ICAM - 1 and P - selectin could be observed in rat pancreas of group M at 3, 6 and 12 hours after operation, and the expression degree increased with time. The expression of both indexes above could also be observed in rat pancreas of group M, but the expression degree decreased with time. Conclusion By reducing the neutrophil infiltration and the expression of ICAM - 1 and P - selectin in rat pancreas, baiealin can bring protective effects on pancreatic tissue in rat model of SAP.
