异氟醚对大鼠脑缺血/再灌注诱导c-Jun磷酸化的影响

摘  要:目的观察大鼠脑缺血/再灌注后c-Jun蛋白磷酸化情况以及异氟醚对其的影响。方法四动脉结扎法建立大鼠脑缺血模型。随机分为假手术组、直接脑缺血/再灌注组、吸入2 h 1.5 MAC异氟醚脑缺血/再灌注组和吸入纯氧2 h脑缺血/再灌注组,全脑缺血15 min后分别再灌注6 h。再灌注6 h的大鼠进行磷酸化c-Jun(p-c-Jun)的免疫印迹和免疫组化检测。结果缺血前吸入1.5 MAC异氟醚组大鼠的c-Jun磷酸化水平明显低于直接脑缺血/再灌注组和吸入纯氧2 h脑缺血/再灌注组(P<0.05)。结论异氟醚抑制c-Jun蛋白的磷酸化可能是其对大鼠缺血/再灌注性脑损伤产生保护作用的机制之一。Objective To observe the focal cerebral ischemia/reperfusion - induced phosphorylation of c - Jun in rats and investigate the effect of isoflurane. Methods Animal model of global cerebral ischemia was established by ligation of bilateral common carotid and vertebral arteries ( four vessels). Healthy male Sprague - Dawley rats were randomly divided into four groups: sham operation group (S) , ischemia/reperfusion group (I/R) , 2 h 1.5 MAC isoflurane preconditioning + ischemia/reperfusion group ( I/R + Iso) and 2 h pure O2 inhalation + ischemia/reperfusion group ( I/R + O2 ). 15 rain after global cerebral ischemia, the rats were allowed 6 hours of reperfusion. Determination of phosphoryla- tion of c - Jun was performed by Western blotting and immunohistochemistry. Results The phosphorylation levels of c - Jun in I/R + Iso group was markedly lower than that in I/R and I/R + O: groups ( P 〈 0.05 ). Conclusion Isoflurane is able to inhibit the phosphorylation of c - Jun, which might be one of its protective mechanisms in the ischemia/reperfusion -induced brain injuries in rats.
