摘  要:目的:探讨超氧化物歧化酶(SOD)对肝癌细胞增殖的抑制作用。方法:利用逆转录病毒载体介导的基因转移技术,将人铜锌SOD基因导入人肝癌细胞系HepG2细胞内,获得高表达。检测其对HepG2细胞生物学特性的影响。结果:与对照相比SOD基因转染后的肝癌细胞生长受到抑制,细胞体积变大,分裂象减少,S期细胞比率减少,软琼脂集落形成率低,裸鼠成瘤瘤体小。结论:铜锌SOD基因具有抑制HepG2肝癌细胞增殖的作用。Objective: To explore the effect of superoxide dismutase (SOD)on inhibition of HepG2 hepatocellular cancer cell proliferation. Methods:Using gene transfer technique, hepatocellular cancer cells(HepG2) were transfected with retroviral vector containing human CuZnSOD cDNA and the elevated SOD gene expression was compared with the parental and neo control cells. The biological characteristics of the cells were measured. Results:Compared with the control cells, cancer cells transfected with SOD gene showed an inhibited growth rate,an increased cell mass, a reduced number of cells in S phase and a decreased clone forming ability in soft agar as well as a smaller tumor size formed in nude mice. Conclusion: The CuZnSOD gene overexpression could inhibit the proliferation of HepG2 hepatocellular cancer cells.
