摘　　要：目的:研究中国汉族人P450-2D6酶基因Ch多态性对血浆利培酮和9-OH利培酮浓度的影响。方法:30例精神分裂症患者应用利培酮达治疗量1周后,用聚合酶链反应-限制性片段长度多态性分析法(PCR-RFLP)分析患者P450-2D6酶基因Ch多态性。测定患者的血浆利培酮、9-OH利培酮浓度。结果:30例精神分裂症患者中P450-2D6酶基因为Ch/Ch型者7例(23.3%),W/Ch型者15例(50.0%),W/W型者8例(26.7%)。W/W型患者平均血浆利培酮浓度(5.28±1.61)ng/ml,血浆9-OH利培酮浓度(17.13±8.15)ng/ml,W/Ch型患者分别为(5.00±2.10)ng/ml及(15.29±6.74)ng/ml,Ch/Ch型患者分别为(5.89±3.34)ng/ml及(15.97±10.16)ng/ml,三组患者两两间血浆利培酮浓度、9-OH利培酮浓度比较差异均无显著性(P均>0.05)。结论:450-2D6酶基因Ch多态性对血浆利培酮浓度、9-OH利培酮浓度无显著影响。Objective： To study the Ch polymorphism of cytochrome p450-2D6 gene in Chinese Han people and its relationship with plasma risperidone and 9-OH risperidone level. Method：30 patients who met the schizophrenia criteria of Chinese classification and diagnostic criteria for mental disorders （ 3rd edition ） （ CC-MD-3 ） were treated with risperidone for one week. After one week treatment Ch polymorphism of cytochrome p450-2D6 gene was analyzed with PCR-RELP method, and plasma risperidone and 9-OH risperidone level were measured. Results：Of the 30 patients, there are 7 Ch/Ch homozygotes （23.3%）, 15 W/Ch homozygotes （50.0%） , 8 W/W heterozygotes （26.7%）in p450-2D6 gene. The plasma risperidone, 9-OH risperidone level was （5.28 ± 1.61 ） ng/ml, （ 17.13 ± 8. 15 ） ng/ml respectively in 8 W/W homozygote patients, The plasma risperidone, 9-OH risperidone level was （5.00 ±2.10） ng/ml, （15.29 ±6.74） ng/ml respectively in 15 W/ Ch homozygote patients,The plasma risperidone ,9-OH risperidone level was （5.89 ± 3.34） ng/ml, （15.97 ± 10.16） ng/ml respectively in 7Ch/Ch heterozygote patients. There wasn＇t significant difference in plasma ris- peridone and 9-OH risperidone among the three types of patients. Conclusion： The polymorphism of cytochrome p450-2D6 gene in Ch has no significant influence on plasma risperidone and 9-OH risperidone level in schizophrenic patients treated with risperidone.
