摘 要:目的探讨p16基因启动子异常甲基化在肺癌分子诊断中的应用价值。方法收集肺癌患者的血浆,提取游离DNA,经亚硫酸氢钠修饰并回收纯化,进行甲基化特异性PCR(MSP)。结果51例患者血浆中有32例检测到p16基因启动子发生了异常甲基化,检出率为62.7%,对照血浆均为阴性。结论血浆p16基因甲基化检测可能有助于肺癌的早期诊断。Objective To investigate the p16 gene promoter aberrant methylation in the plasma of patients with lung cancer and its possible clinical application in molecular diagnosis of lung cancer. Methods Free DNA was extracted from peripheral blood plasma of lung cancer patients. After modifying with sodium bisulfite and purifying, then they were used as template for methylation specific PCR (MSP). Results p16 gene promoter aberrant methylation was found in 32 of 51 samples of lung cancer patients (62.7%) using methylation specific PeR,no methylated p16 gene was detected in control samples. Conclusions The measurement of methylated p16 gene may be helpful in early diagnosis of lung cancer.
