摘 要:[目的]研究杜仲内生真菌的抗氧化活性和菌种鉴定。[方法]采用组织分离法,筛选杜仲叶片的内生菌种。菌株No.173的总抗氧化活性最高,作为供试菌种。制备它的发酵液提取物,测定了提取物抗氧化能力。对菌株No.173进行了形态和分子鉴定。[结果]处理4 d后,处理组的POV值极显著性低于对照。第6天起,处理组的效果明显好于阳性对照组(0.04%Vc和VE)。菌株No.173在PDA平板上的菌落初期为白色,渐变至暗青色,背面褐色,菌落边缘不整齐。序列比对结果显示,菌株No.173 rDNA ITS扩增片段的碱基序列与球毛壳菌序列相似性为100%。[结论]杜仲叶片中内生真菌菌株No.173为球毛壳菌,具有明显和稳定的抗氧化活性作用。[Objective] The aim was to study the antioxidant activity and strain identification of an endophytic fungal from Eucommia ulmoides Oliv.[Method] The tissue separation method was used to screen the endophytic fungi in the leaves of E.ulmoides.Strain No.173 showed highest antioxidant activity and could be used as tested strain.Its broth extract was prepared to determine its antioxidant activity. Strain No.173 was identified from morphology and molecule.[Result] After treatment for 4 d,the POV value of treatment group was significantly lower than that of control group.From 6th d,the effect of treatment group was better than that of positive control group(0.04%Vc and VE).The colony of strain No.173 was white at the primary stage on the PDA plate and gradually changed to dark blue.Its back was brown and the edge of colony was irregular.The result of sequence alignment showed that the similarity between the base sequence of rDNA-ITS amplified fragment of strain No.173 and that of Chaetomium globosum Kunze was 100%.[Conclusion] Endophytic fungi strain No.173 in E.ulmoides leaves belonged to C.globosum and had significant and stable antioxidant activity.
