摘 要:目的探讨α-连接素(α-catenin)在进展期胃癌转移淋巴结内癌细胞的再表达情况及其临床意义。方法采用免疫组化、蛋白印记等方法检测30例胃癌患者胃癌原发灶和转移淋巴结中α-连接素蛋白表达情况。结果在30例胃癌原发灶中100%(30/30)α-连接素表达程度减低,胃癌转移淋巴结中50%(15/30)再表达阳性,其中高分化腺癌的再表达阳性率为71.42%(10/14);中低分化腺癌的再表达阳性率为31.25%(5/16)。高分化腺癌相对于中低分化腺癌更易出现α-连接素的再表达阳性,两者比较差异具有统计学意义(P〈0.05)。30例原发肿瘤组织及相应转移淋巴结均检测到不同程度的α-连接素蛋白表达。原发癌组织中的各例蛋白相对强度为11~170,平均105±42,转移癌淋巴结组织中各例蛋白相对强度为8~112,平均40±28,原发癌组织中α-连接素蛋白表达相对强度高于转移淋巴结组织。在转移淋巴结中高分化腺癌蛋白表达相对强度为25~112,平均54±33,中低分化腺癌的蛋白表达相对强度为8~55,平均27±12。两者间蛋白表达相对强度差异具有统计学意义(P〈0.01)。结论胃癌转移淋巴结内癌细胞是存在α-连接素的再表达的,其蛋白再表达水平与肿瘤分化程度相关,并且对癌细胞在转移灶内聚集再生长起着重要作用。Objective To investigate the significance of re-expression of α-catenin in lymph nodes of advanced gastric cancer metastases. Me, otis The expression of α-catenin was detected in 30 patients with gastric cancer by immunohistochemistry and protein imprinting. Results The expression of α-catenin in primary focus of 30 patients was decreased( 100% ), while which was positive(50% ) in metastatic advanced gastric cancer, in which the positive rate in well-differentiated adenocarcinoma was 71.42% (10/14), which was 31. 25% (5/16) in poorly differentiated adenocarcinoma. Compared with poorly differentiated adenocarcinoma, the well-differentiated adenocarcinoma was apt to positive of α-catenin expression, there was a significant difference bewteen the two different differentiated adenocarcinoma ( P 〈 0.05). The positive expression of α-catenin was found both in primary tumor tissue and corresponding metastatic lymph nodes. The relative intensity of protein in primary carcinoma was 11 - 170 (mean 105±42), which in lymph nodes of metastatic cancer was 8 - 112 (40±28), the relative intensity of expression of α-catenin in primary carcinoma was higher than that of metastatic lymph node. The relative intensity of expression of α-cateninIn in well-differentiated adenocareinoma of metastatic lymph nodes was 25 - 112 (mean 54±33), which was 8 - 55 (mean 27 ± 12) in poorly differentiated adenocarcinoma. There was a significant difference in relative intensity of expression of α-eatenin between the two different differentiated adenocarcinoma. Conclusion The re-expression of α-catenin exists in lymph nodes of metastatic advanced gastrie cancer, and the re-expresian levels are related with the extent of tumor differentiation, furthermore, which plays an important role in gathering and growth of cancer cells in metastatic focus.
