摘  要:目的探讨1,25二羟维生素D3[1,25(OH)2D3]对胆管癌细胞系QBC939的体外增殖及凋亡的影响。方法将不同浓度的1,25(OH)2D3与胆管癌细胞系QBC939共同培养,采用MTT法测定细胞增殖能力、显微镜观察细胞形态学的改变、流式细胞仪检测细胞周期与凋亡、免疫细胞化学观察bcl-2的表达。结果0.1~0.5μmol/L的1,25(OH)2D3对胆管癌细胞QBC939有抑制作用,呈剂量依赖性。经1,25(OH)2D3作用72h后细胞G1期比例升高,S期比例下降,其中0.5μmol/L组细胞G1期由(50.3±1.0)%上升至(65.5±3.2)%,S期由(39.4±0.5)%下降至(23.6±0.7)%;并且可诱导细胞产生凋亡,0.5μmol/L组作用后细胞凋亡率由0.5%上升至24.6%;bcl-2的表达下调。结论1,25(OH)2D3能抑制胆管癌细胞系QBC939增殖并诱导细胞凋亡,其引起凋亡的机制可能与下调bcl-2的表达相关。Objective To explore the effect of 1,25(OH)2D3 on the growth and apoptosis of cholangioearcinoma cell line QBC939 in vitro. Methods Cholangioearcinoma cell line QBC939 was treated with different concentrations of 1,25 (OH) 2D3. Cell proliferation was tested by MTT assay ,morphologic changes were observed under a light microscope, cell cycle and apoptosis were analyzed by flow cytometry (FCM), and immunocytochemistry was used to detect the expression of bcl -2. Results 1,25 (OH)2D3 inhibited the proliferation of cholangiocarcinoma ceil line QBC939 at a concentration range of 0.1- 0.5μmol/L in a dose - dependent manner. After treated with 1,25 (OH) 2 D3 for 72 h, the percentage of cells in G1 phase was significantly increased, and that in S phase decreased. 0.5μmoL/L 1,25(OH)2D3 increased the percentage of cells in GI phase ceils from (50.3 ± 1.0)% to (65.5 ± 3.2) %, and decreased that in S phase from ( 39.4 ±0.5 ) % to ( 23.6 ± 0.7 ) %. 1,25 (OH) 2 D3 could also induce apoptosis, and 0.5 μmol/L 1,25 ( OH )2 D3 increased the apoptosis rate from 0.5 % to 24.6%. The expression of bel - 2 was decreased. Conchlsion 1 , 25 (OH) 2 D3 can inhibit the proliferation and induce the apoptosis of cholangioearcinoma cell line QBC939, which may be related to the down - regulation of the bcl - 2 expression.
