摘 要:目的观察丹参酮ⅡA对人肝癌SMMC-7721细胞的增殖抑制和凋亡诱导作用,及对血管内皮生长因子(VEGF)表达的影响,探讨其可能的抗肿瘤作用机制。方法体外培养人肝癌SMMC-7721细胞株,MTT比色法观察细胞增殖抑制情况;荧光显微镜检测细胞凋亡;免疫细胞化学法和ELISA法分别检测细胞及细胞培养液中VEGF的表达情况。结果丹参酮ⅡA对肝癌细胞的生长有明显的抑制作用,并呈剂量依赖性,以0.5mg/L终质量浓度的抑制作用最明显,其48h的抑制率为72.5%,与未加药对照组相比差异具有统计学意义(P<0.01);荧光显微镜下经丹参酮ⅡA作用后的肝癌细胞可见凋亡小体形成;免疫细胞化学染色及ELISA法检测发现,丹参酮IIA作用组肝癌细胞VEGF表达和培养液中VEGF分泌量明显减少,与未加药对照组相比差异有统计学意义(P<0.01)。结论丹参酮ⅡA可下调细胞VEGF的表达,这可能是其抗肿瘤作用的机制之一。Objective To evaluate the effects of Tanshinone IIA in inhibiting the proliferation, inducing apoptosis and the expression of vascular endothelial growth factor (VEGF) on SMMC-7721 cell and to investigate the possible antitumor mechanism of Tanshinone IIA. Methods The human hepatocellular carcinoma cell line SMMC- 7721 was cultured in vitro. The cell growth activity was evaluated by MTT assay. The apoptosis of SMMC-7721 was studied through the fluorescence microscope. The expression of VEGF of SMMC-7721 cell and the cell culture fluid was determined by immunocytochemistry and ELISA assay, respectively. Results Tanshinone IIA treatment of SMMC-7721 cells resulted in a strong growth inhibition in a dose-dependent manner. The peak value of inhibition rate (72.5%) reached at 0.5 mg/L Tanshinone IIA for 48h, and there were significant differences compared to control group (P〈0.01). The formation of apoptotic bodies of SMMC-7721 cell was observed in Tanshinone IIA-treated group under the fluorescence microscope. The expression of VEGF in Tanshinone IIA-treated group was significantly lower than that in control group (P〈0.01). The similar result could be observed in cell culture fluid with ELISA assay. Conclusion Tanshinone IIA can inhibit the proliferation and induce the apoptosis of SMMC-7721, and these results may be related to the down-regulation of VEGF.
