摘 要:目的:分析睡眠剥夺条件下大鼠中枢神经系统c-fos和nestin在不同部位的表达特点,为进一步研究睡眠剥夺所造成生理影响的分子机制提供线索.方法:采用小平台水环境法建立大鼠睡眠剥夺24,48,72h模型,设立大平台对照组和正常饲养对照组,RT-PCR方法分别检测各组大鼠海马、皮质和小脑中c-fos和nestin的表达水平.结果:与对照组相比,睡眠剥夺组c-fos表达升高,nestin表达降低.结论:睡眠剥夺影响大鼠中枢神经系统c-fos和nestin基因的表达.AIM: To analyze the expression characteristics of c-fos and nestin in the different parts of rat central nervous system after sleep deprivation(SD) in different times and to provide clues on the molecular mechanism of sleep deprivation. METHODS: SD was induced by housing rats on the small platforms over water. The rats in control group were housed in the normal cage or on the large platforms over water. The expression level of c-fos and nestin in hippocampus, cerebellum and cortex was examined at 24, 48 and 72 h after sleep deprivation by RT-PCR. RESULTS: c-fos was up-regulated and nestin was down-regulated in SD models compared with control. CONCLUSION: Under sleep deprivation condition, the expression level of c-fos and nestin is regulated in rat CNS.
