朝鲜族人群DXS 9898基因座遗传多态性研究

摘　　要：[目的]探讨延边地区朝鲜族群体短串联重复序列(STR)基因座DXS 9898的遗传多态性分布,获取相应多态性的群体遗传学数据.[方法]随机抽取294名延边地区无血缘关系的朝鲜族个体,其中男性为187名,女性为107名,采集静脉血,提取DNA,行PCR扩增,变性聚丙烯酰胺凝胶电泳及银染显色,行遗传基因多态性分析.[结果]在DXS 9898基因座中检测出7个等位基因和15个基因型,基因型频率分布符合Hardy-Weinberg平衡定律;杂合度高于0.523 4,多态信息含量高于0.540 0,女性组个人识别力为0.780 0,男性组单倍型多样性为0.610 9.[结论]DXS 9898基因座具有较高的多态信息量和个人识别力,是较理想的遗传标记系统.OBJECTIVE To understand the allele structure and genetic polymorphism at DXS 9898 short tandem repeat （STR） locus in the Korean-Chinese nationality of Yanbian area, and to construct a preliminary database. METHODS The venous blood with no sibship was taken from 294 cases （187 of male, 107 of female） of Korean-Chinese nationality. The DXS 9898 short tandem STR locus was performed by using the techniques of PCR, polyacylamide gel electrophoresis and silver staining. RESULTS Eight allele were observed at DXS 9898 locus, and the distribution of genotypes fitted for the Hardy-Weinberg equilibrium. The statistical analysis of DXS 9898 ocus showed the heterozygosities was more than 0. 523 4, the polymorphic information contents （PIC） was more than 0. 540 0; the power discrimination （PD） and haoplotype diversity （HD） were more than 0. 780 0 and 0. 610 9, respectively. CONCLUSION The DXS 9898 locus in this study are found to have relative high PIC and PD, so DXS 9898 can provide useful markers for genetic purposes.
