摘  要:目的探讨食管鳞癌及正常食管黏膜组织中热休克蛋白70(HSP70)的表达状况及其意义。方法取食管鳞癌手术切除标本33例,液氮保存。提取新鲜组织中的蛋白质western blotting检测HSP70蛋白的表达;RT-PCR方法检测HSP70mRNA的表达,应用ImageTool3.0图像分析软件对检测结果进行定量分析。结果食管鳞癌中HSP70mRNA和蛋白阳性表达率分别为100%和93.9%,与食管正常黏膜相比明显升高(分别为69.7%和45.5%);食管鳞癌组织和正常食管黏膜中的HSP70mRNA的相对表达量分别为(1.35±0.21)和(0.73±0.19),差异有显著性(P<0.01);HSP70蛋白表达与食管鳞癌淋巴结转移相关,与年龄、性别和浸润深度无关。结论食管鳞癌组织中HSP70表达水平明显升高,且与淋巴结转移相关,HSP70基因可能参与了食管鳞癌的发生、发展过程。[Objective] To study the expression and significance of heat shock protein 70 (HSP70) in the normal esophageal mucosa and esophageal squamous cell carcinoma (ESCC). [Methods] Fresh samples were obtained from 33 patients who underwent resection of ESCC. All samples were preserved in liguid nitrogon. The expression of HSP70 protein and HSP70 mRNA were studied using Western Blotting and reverse-transcription polymerase chain reaction (RT-PCR) respectively, followed by quantitative analysis using Image Tool 3.0 analysis software. [Results] The positive rate of HSP70 mRNA and protein in ESCC was 100% and 93.9%, and Was higher than that in normal esophageal mucosa (69.7% and 45.5%, respectively). The relative expression of HSP70 mRNA in ESCC and normal esophageal mucosa was (1.35±0.21) and (0.73±0.19) respectively. The difference between eachother have statistical significance (P 〈0.001). The expression of HSP70 protein did not correlated with patient's age, gender and the depth of tumour infiltration but correlated with lymphonode metastasis. [Conclusion] The expression of HSP70 in ESCC was markedly increased and correlated with the status of metastasis of lymphonode which indicated the expression of HSP70 took part in the carcinogenesis and development of ESCC.
