摘 要:目的探讨少突胶质细胞系基因(01igodendrocyte lineage gene)Olig2和同源盒基因(Homeobox)Hb9在小鼠胚胎脊髓发育过程中的表达情况。方法选择小鼠的9.5~13.5d胚胎脊髓,并分为E9.5d组、E10.5蝴、E12.5d组、E13.5d组;采用免疫荧光方法检测Olig2和Hb9在胎鼠脊髓中的分布,同时用流式细胞仪检测分别标记Olig2和Hb9的阳性细胞百分率。结果免疫荧光染色显示在这5组胎鼠脊髓的腹侧都有Olig2和Hb9的表达,流式细胞仪检测显示E9.5d组Olig2阳性细胞百分率高于其它4组,差异有统计学意义(P<0.05),E11.5d组Hb9的阳性细胞百分率高于其它4组,差异有统计学意义(P<0.05)。结论Olig2和Hb9可能与胎鼠脊髓运动前体细胞和运动神经元的发育有关。Objective To study oligodendrocyte lineage gene Olig2 and Homeobox gene Hb9 expression in the process of Mouse embryonic spinal cord. Methods Mouse embryonic spinal cord was selected from E 9. 5 d to E13.5 d which were divided into a E9. 5 d group, a E10. 5 d group, a Ell. 5 d group, a E12. 5 d group,a E13. 5 d group. The expression of Olig2 and Hb9 were detected with immunohistofluorescence in Mouse embryonic spinal cord, Olig2 and Hb9 positive cells were counted respectively by Flow Cytometry (FCM). Results In spinal cord of five groups there were Olig2 and Hb9 positive with immunohistofluorescence. Flow cytometry showed the rate of Olig2 positive cell in E 9. 5 d group was higher than that in other four groups(P〈0. 05), the rate of Hb9 positive cell in E 11.5 d group was higher than that in other four groups(P〈0.05). Conclusions Olig2 and Hb9 may be concerned with the development of the mouse spine cord motor neuron.
