摘　　要：细胞周期素依赖性激酶-5(cylin-dependent kinase-5,Cdk5)作为细胞周期素依赖性激酶家族成员之一,其生理作用主要表现为影响神经细胞的正常发育和维持有丝分裂后神经细胞的功能及骨架结构。Cdk5催化底物磷酸化功能的失调与一些神经退行性疾病有着密切的关系。该文重点对Cdk5介导的Alzheimer病的病理过程及已报道的Cdk5抑制剂进行综述,探讨该靶点药物开发中存在的问题及可能的解决办法。Cylin-dependent kinase-5 is a member of cylin-dependent kinases family. It significantly participates in regulating neural development and maintaining the function and architecture of adult nerve cell. Deregulation of Cdk5 in phosphorylating its substrates was closely implicated in a series of neurodegenerative diseases. In this paper,the role of Cdk5 in Alzheimer＇s disease and inhibitors reported lately were reviewed, and then the open question and solution in developing drugs on this target were also followed.
