摘  要:目的考察FL-2大孔吸附树脂对山楂总黄酮成分的分离纯化方法。方法根据山楂黄酮的结构特点,考察FL-2树脂的吸附性能,对FL-2吸附树脂纯化山楂总黄酮的工艺条件进行了筛选,并采用紫外-可见分光光度法对山楂总黄酮含量进行定量分析。结果FL-2大孔吸附树脂对山楂中总黄酮具有较好的吸附选择性。吸附纯化的最佳工艺条件为吸附流速1BV/h,以60%乙醇作为洗脱剂,洗脱流速为2BV/h。采用上述工艺条件,山楂总黄酮的得率为4.78%,产品纯度达到68.71%。结论FL-2树脂用于山楂总黄酮的分离纯化简单有效,易于工业化推广。Objective To study the method for separating and purifying the total flavonoids in Crataegus pinnatifida Bge. (TFC) with macroporous adsorption resin FL-2. Methods According to the structural characteristic of flavone, the resin FL-2 was systematically studied for its adsorption capability. The technological conditions of the adsorption resin FL-2 for separation of TFC was selected. UV-Vis was used to measure the content of TFC. Results The macroporous adsorption resin FL-2 can be used to produce the TFC with high quality. The optimal technological conditions were: the flow velocity of adsorption, 1 BV/h; the eluting reagent, 60% ethanol; eluting velocity, 2 BV/h. The purity and yield of the TFC were 68.71% and 4.78% with the technological conditions respectively. Conclusion It is a simple and efficient method to separate the total flavonoids from Crataegus pinnatifida Bge. by using FL-2 resin, and can be applied to manufacture.
