抑癌基因PTEN与食管鳞癌血管新生及预后相关性研究

摘　　要：[目的]探讨食管鳞状细胞癌(ESCC)抑癌基因PTEN蛋白表达及其对血管新生和预后的影响。[方法]免疫组化SP法检测102例ESCC组织及其相应的30例癌旁正常组织中PTEN蛋白表达,检测ESCC组织微血管密度(MVD),Kaplan-Meier法分析与预后的关系。[结果]ESCC中PTEN阳性表达率为63.7%(65/102),在癌旁正常食管组织中达到96.7%(29/30)(P<0.01)。PTEN表达强度与浸润深度及是否伴淋巴结转移明显相关(P<0.05),与MVD呈负相关(P<0.01),而与ESCC预后呈正相关。[结论]PTEN表达下调可能参与ESCC发生及肿瘤血管生成,有望成为评价ESCC患者预后的较好指标。[Purpose] To explore the protein expression of PTEN in esophageal squamous cell carcinoma（ESCC） and its effect on ESCC angiogenesis and prognosis. [Methodsl The protein expressions of PTEN in 102 cases with ESCC specimens and 30 cases with pericarcinoma esophageal tissues were examined by S-P immunobistochemical staining. MVD was detected with anti-CD34 monoclonal antibody and its relationship with prognosis was analyzed by Kaplan-Meier life table method. [Results] The positive rate of PTEN in ESCC was 63.7%（65/102） and in the pericarcinoma esophageal tissues was 96.7%（29/30）（P〈0.01）.The expression of PTEN significantly related to lymph node metastasis and depth of tumor invasion （P〈0.05）. The expression intensity of PTEN showed negative correlation with MVD（P〈0.01） and positive correlation with prognosis. [Conclusion] Down regulation of PTEN expression might play an important role in tumorigenesis, tumor angiogenesis. PTEN might become a prognostic marker for appreciating prognosis of ESCC in the future.
