摘 要:目的内皮细胞(EC)与平滑肌细胞(SMC)相互作用是动脉粥样硬化的重要病理生理特征,而内皮脂肪酶(EL)是内皮细胞在受致动脉粥样硬化因素作用后高表达的一种酶。目前尚无关于内皮细胞表达EL后对平滑肌细胞的生物学特性影响报道。方法通过质粒转染法使脐静脉内皮细胞(HUVEC)高表达EL(简称HUVEC-EL),并与正常HUVEC对照,采用共培养技术观察HUVEC高表达EL后对内皮细胞的增殖、迁移、EL表达的影响。结果内皮细胞高表达EL后,可促进共培养的平滑肌细胞的增殖、迁移,但平滑肌细胞所表达EL量减弱。结论内皮细胞高表达EL后对共培养平滑肌细胞的生物学活性产生影响,这些影响可能参与动脉粥样硬化的病理生理过程。Objective The interaction between endothelial cells and smooth muscle cells(SMCs) is a significant pathophysiological characteristics of atherosclerosis. Endothelial lipase(EL) is overexpressed on endothelial cells exposed to proatherosclerotie factors. But there is no report about the effect of EC over-expressing EL on the bionomics of co-cultured SMCs Methods The HU-VEC over-expressing EL (named HUVEC--EL) was got through transfection HUVEC with plasmid pcDNA4-EL-myc/His. Compared with the nomal HUVEC,the influence of HUVEC over-expressing EL on the proliferation, migration and the expression of EL on the co-cultured SMCs were evaluated. Results After HUVEC over-expressing EL,the ability and the speed of proliferation of SMCs was enhanced,and the number of migration was increased. But the expression of EL was decreased. Conclusion After HUVEC over-expression of EL, the bionomies of co-cultured SMCs is changed,and these changes may participate the pathophysiology of atherosclerosis.
