软骨肉瘤细胞Caspase-3表达及其与相关基因相互作用的研究

摘　　要：目的:研究肿瘤基因Caspase-3与PTHrP和Sox9在人体软骨肉瘤细胞HTB-94中的相互作用机制。方法:小干扰siRNA分别阻断PTHrP和Sox9后,检测肿瘤细胞中Caspase-3的mRNA和蛋白表达。结果:PTHrP基因被干扰后,Caspase-3的mRNA表达增加了57.7%,蛋白的表达增加了57.1%;Sox9基因被干扰之后,Caspase-3的mRNA表达增加了47.4%,蛋白的表达增加了48.2%。结论:肿瘤基因Caspase-3在软骨肉瘤细胞中的表达与软骨肉瘤相关基因PTHrP和Sox9有关,可能是位于PTHrP和Sox9的下游基因。OBJECTIVE： Tostudy the correlated function- al mechanism between Caspase-3 and PTHrp, Sox9 genes re- spectively in chondrosarcoma cell line. METHODS： The Caspase-3 mRNA and protein expression were tested after siRNA interfered the genes of PTHrp and Sox9. RESULTS： After interfering PTHrP the mRNA expression of Caspase-3 was in- creased by 57.7%, and the protein expression was increased by 57.1% ; after interfering Sox9 the mRNA expression of Caspase-3 was increased by 47.4% , and the protein expression was increased by 48.20%. CONCLUSIONS： The inhibition of PTHrp and Sox9 can inhibit the expression of Caspase-3 gene. Caspase-3 may be the down stream gene in the rest of PTHrP and Sox9 genes.
