摘  要:本文采用正交试验设计优选ConA 诱导的人外周血淋巴细胞化学发光(PBL—CL)测定体系中PBL、Luminol 及ConA 等三个主要因素的最佳组合。结果表明,PBL—CL 测定的最优实验条件为:0.5ml PBL 悬液(1×10~6/ml)、0.2ml Luminol 溶液(5×10^(-4)M)及0.2ml ConA 溶液(500μg/ml),反应液总量为1.0m。PBL—CL 测定是一种简便、怏速、敏感的检测淋巴细胞早期活化及功能的新方法。Lymphocyte chemiluminescence is one of the early events involved in the activation of lymphocytes.This CL is thought to result from the generation of oxygen radicals (OO(?),H_2O_2,.OH,~1O_2)by the lymphocytes,the consequent oxi- dation of luminol and emissin of light.In this paper,the experimental condition for ConA-induced human peripheral blood lymphocyte chemiluminescence (PBL-CL) was investigated by using orthogonal experiment design.The kinetics of CL respo nse in human PBL to ConA was also studied.The results showed that the optimal combination of three essential factors in PBL-CL assay was 0.5ml PBL suspension (1×10~8/ml),0.2ml luminol solution (5×10~^(-4)M) and 0.2ml ConA solution (500 μg/ml),The total volume is 1.0ml. PBL—CL assay might serve as a simple,rapid and sensitive method to evaluate the early activation and function of lymphocytes.
