摘 要:目的观察不同浓度的维生素K3(VK3)对人膀胱癌细胞株T24凋亡的诱导作用。方法采用噻唑蓝(MTT)法观察不同浓度VK3(0,2,5,10,20μmol/L)对T24细胞增殖的影响,采用流式细胞术检测不同浓度的VK3对细胞凋亡率的影响。结果0,2,5,10,20μmol/LVK3作用T24细胞48h后,生长抑制率分别为(5.80±0.038)%,(13.30±0.036)%,(26.55±0.032)%,(42.58±0.028)%和(65.35±0.034)%,细胞凋亡率分别为(2.86±0.4)%,(5.16±0.7)%,(10.85±1.1)%,(23.01±0.8)%和(75.80±1.2)%,其中10μmol/L,20μmol/LVK3对T24细胞生长有明显抑制作用(P<0.05)。结论10-20μmol/LVK3在体外作用48h后可明显地诱导膀胱癌T24细胞株发生凋亡。Objective To explore the inducing effect of vitamin K3 on apoptosis of human bladder cancer cell line T24. Methods T24 cells were cultured in the presence of different concentrations of VK3 (0,2,5,10,20 μ mol/L). The inhibitory rates of VK3 were detected by MTF colorimetry,and apoptosis was observed using flow cytometry. Results After T24 cells were cultured with VK3 for 48 h at concentrations of 0,2,5,10,20 μmol/L, the inhibitory rates of VK3 were (5.80 ± 0. 038 ) %, ( 13.30 ± 0. 036) %, (26.55 ±0.032) %, (42.58 ± 0. 028) % and (65.35 ± 0.034) %, respectively,and the apoptotic rates were(2.86 ± 0.4) %, (5.16 ±0.7) %, ( 10.85 ± 1.1 ) %, (23.01±0.8 ) % and ( 75.80± 1.2 ) %, respectively. Cell growth significantly decreased in the presence of 10 μmol/L,20 μmol/LVK3 ( P 〈 0.05). Conclusion VK3 at the concentrations of 10 - 20 μmol/L could obviously inhibit the proliferation of T24 ceils and induce apoptosis after 48 h culture in vitro.
