摘 要:目的:探讨EB病毒膜抗原(MA)的表达与系统性红斑狼疮的关系。方法:用直接免疫荧光抗体法(DFA)检测MA在转基因小鼠体内的表达,用间接免疫荧光法(IFA)和免疫印迹法(IBT)分别检测血清中的抗核抗体(ANA)和抗ENA抗体,用Bradford法检测小鼠24h尿蛋白浓度。常规病理HE染色观察小鼠的肾脏病理改变,冰冻切片免疫荧光检测肾脏是否有免疫复合物沉积。结果:表达MA的转基因小鼠组,有8只小鼠ANA阳性,5只小鼠抗ENA抗体阳性,24h尿蛋白浓度平均值为4153.8 g/ml,小鼠肾脏存在多种病理改变及大量免疫复合物在肾小球沉积。对照组小鼠ANA、抗ENA抗体均为阴性,24h尿蛋白浓度平均值为382.7g/ml,肾脏未见病理改变。结论:EB病毒MA的表达可能引发SLE。Objective: To find the relationship between the expression of Epatein-Barr virus membrane antigen (MA) and systemic lupus erythematosus (SLE). Methods: The expression of MA in BLLFl-transgenic mouse was dectected through the direct immunofluo-rescence antibody technique (FDA). Indirect immunofluorescence antibody (IFA) and immunoblotting technique (IBT) were applied respectively to detect anti-nuclear antibody (ANA) and anti-ENA antibody. Bradford technique was used to measure 24 hours urine of the mice. Pathologic changes of the mouse kidney was observed by HE dyeing. Results: In the experimental group expressed MA, eight mice's blood serum had ANA and five had ENA. The average concentration of the 24h urine protein was 4153.8 g/ml. There were many pathological changes and much immune complex found in the kidneys. Whereas in the control group, there was no MA, no ANA and ENA in the blood serum of the mice, and the kidneys were normal. The average concentration of the 24h urine protein was 382.7 g/ml. Conclusion: The expression of MA may induce the SLE.
