摘 要:目的研究亚高温盆腔区域热疗能否诱导原位的大鼠浸润性膀胱肿瘤热休克蛋白70(HSP70)表达增加。方法制备原位的大鼠浸润性膀胱肿瘤动物模型,分为对照组(A组)、单次热疗组(B组)、间隔24 h再次热疗组(C组)、间隔72 h再次热疗组(D组)。从体外加热大鼠盆腔至41℃,持续1 h。用胶体金免疫电镜、免疫组化、蛋白芯片检测肿瘤细胞HSP70表达。结果A组HSP70定位于肿瘤细胞质,B、C、D组HSP70均定位于肿瘤细胞核、线粒体和细胞质。B、C、D组HSP70阳性表达率均高于A组,差异有统计学意义(P<0.01),但B、C、D组组间差异无统计学意义(P=0.101)。B、C、D组HSP70表达量高于A组,差异有统计学意义(P<0.01);C、D组HSP70表达量均高于B组,差异有统计学意义(P<0.01);C组HSP70表达量高于D组,差异有统计学意义(P<0.01)。结论不同方案的亚高温盆腔区域热疗均能诱导原位的大鼠浸润性膀胱肿瘤发生热休克反应,上调HSP70表达;亚高温盆腔区域热疗是一种温和的加热方法,它不能使全部肿瘤细胞发生热休克反应;HSP70表达率与热疗次数无关,但表达量与热疗次数呈正相关;间隔24 h再次热疗的HSP70表达量最高。Objective To explore whether heat shock protein 70 of rat bladder tumor can be provoked and up-regulated by non-invasive pelvic cavity regional moderate hyperthermia. Methods Six-week-old female inbred line F344 rats were instilled MNU transurethrally once every other week for 6 times. Four weeks later, NRL-00II Double RF Tumor Hyperthermia System was applied to rats. The rat pelvic was heated to 41℃ and persisted for 1 hour. Grouping was as follows (each n= 5) :group A was control,group B was heated once,group C was heated twice interval 24 h,group D was heated twice interval 72 h. The localization of HSP70 of the bladder tumor cells were detected by protein A-10 nm colloidal gold labeled immune electronmicroscopy. The positive cell rates of HSP70 were detected by immunohistochemistry to paraffin embeded bladder tumor. Half-quantified of HSP70 from supernatant of tumor lysate of every specimen was detected by SELDI-TOF-MS. Results HSP70 of group A was located in cytoplasm without heat stress. HSP70 of group B,C and D were located in tumor cell nucleus, mitochondrion and cytoplasm after heat stress, Immunohistochemistry showed that the positive cell rate of HSP70 of group A was significant less than that of group B,C and D(P〈0. 01). Nevertheless there was no significant difference among groups B,C and D(P〈0. 01). SELDI-TOF-MS indicated that quantity of HSP70 were group C, D, B and A in de scending(P〈0. 01). Conclusions The positive rate of HSP70 was not increased with heat times. HSP70 can be provoked and up-regulated by non-invasive pelvic cavity regional moderate hyperthermia in rat bladder tumor, the positive cell rate of HSP70 was not increasing with heating. But the quantity of HSP70 was increasing with heating times. The quantity of HSP70 of group of heated twice interval 24 h was the highest.
