摘  要:本实验采用黑曲霉发酵产β-糖苷酶。应用单因素实验确定最佳碳源为玉米芯,最佳氮源为酵母粉.然后用Plackett—Burman实验设计,筛选出两个显著影响的因素:玉米芯,酵母粉。再用响应曲面法对主要影响因子进行各因子水平及其交互作用优化与评价,并利用SAS软件对该模型进行求解。当各因素分别为玉米芯21.35g/L,酵母粉12.5g/L,pH5.0,KH2PO42g/L,MgSO40.3g/L,CaCl20.3g/L,微量元素:FeSO45mg/L,MnSO41.6mg/L,ZnSO41.0mg/L,发酵液酶活为2.46 U/mL,比原来提高50.92%。The Aspergillus niger was used to produce β-glucosidase . The result of single factor experiment showed that the optimal carbon source was corn core powder and the optimal nitrogen source was yeast extract . Plackett- Burman design was undertaken to evaluate the effect of seven factors. By the statistical regression analysis , the significant factors affecting β-glucosidase activity were determined as follows: corn core powder, yeast extract . Response surface methodology was used to optimize and evaluate the above critical factor and their interaction . It was shown that the optimal concertrations of variables were: 21.75 g/L corn core powder, 12.5g/L yeast extract, pH 5, KH2PO4 2g/L ,MgSO4 0.3g/L ,CaCl2 0.3g/L ,microelement:FeSO4 5mg/L, MnSO4 1.6mg/L, ZnSO4 1.0mg/L. At the optimal fermentation condition the β-glucosidase activity in fermentation liquor was 2.46 U/mL,increased by 50.92%.
