摘  要:目的:通过检测S100A4、基质金属蛋白酶2(matrix metalloproteinase 2,MMP-2)在胃癌组织中的表达,探讨其在胃癌转移中的作用。方法:应用免疫组织化学方法检测S100A4和MMP-2在正常胃黏膜(n=20)、胃癌组织(n=49)中的表达,比较二者在两种组织中的表达差异。结果:S100A4、MMP-2在胃癌组织中的阳性表达率分别为59.18%、67.35%,其染色强度随浸润深度、淋巴结转移和肿瘤分期的增加而增强(P<0.05);在胃癌的不同病理类型和分化程度亚组,S100A4和MMP-2的表达差异均无统计学意义(P>0.05);S100A4在正常胃黏膜组织不表达,MMP-2在正常胃黏膜组织中的表达率为20.00%,两者的表达率均显著低于胃癌组织中的表达率(P<0.01)。S100A4和MMP-2在胃癌组织中的表达呈正相关(r=0.661,P<0.001)。结论:S100A4、MMP-2在肿瘤侵袭转移中发挥了重要作用,联合检测S100A4和MMP-2的表达可用于评定胃癌的转移潜能。Objective:To investigate the expressions of S100A4 and MMP- 2 and their correlations to invasiveness and metastasis in gastric cancer. Methods:The expressions of S100A4 and MMP- 2 were detected by SP immunohistochemical method in 49 cases of gastric cancer tissue and 20 cases of normal tissue. Results:The positive rate of S100A4 and MMP- 2 (59.18% 、67.35%) was significantly higher in gastric cancer tissue than in normal gastric tissue (0.00% 、20.00% ,P〈0. 001). The intensity of S100A4 and MMP- 2 staining in tumor cells was correlated significantly with depth of tumor infiltration(T- stage), lymph node metastasis(N- stage)and TNM stage. No significant correlations were found between S100A4 and MMP- 2 with tumor differentiation and histologic types.There was a positive relationship between the expression of S100A4 and MMP- 2(r=0.661, P〈0.001). Conclusion: S100A4 and MMP- 2 proteins play a critical role in tumor metastasis. Codetection of the expression of S100A4 and MMP- 2 can be used to evaluate the metastatic potential of gastric cancer.
