摘 要:目的:探讨季德胜蛇药对大鼠急性肝损伤的保护作用及其机制。方法:SD大鼠48只,随机分为正常对照组、模型组、高剂量蛇药治疗组和低剂量蛇药治疗组。采用α-萘异硫氰酸酯(Alpha-naphthylisothiocyanate,ANIT)制备大鼠急性肝损伤模型,观察季德胜蛇药对肝损伤大鼠的血清丙氨酸转氨酶(ALT)、门冬氨酸转氨酶(AST)、总胆红素(TBil)及丙二醛(MDA)、超氧化物歧化酶(SOD)、还原型谷光甘肽(GSH)、一氧化氮(NO)水平的影响,并对肝脏组织进行病理学观察。结果:与模型组比较,低剂量蛇药治疗组大鼠的血清ALT水平明显下降而血清GSH显著升高(P<0.05);蛇药治疗组大鼠的肝组织病理改变较模型组减轻,尤以低剂量组明显。结论:季德胜蛇药对ANIT所致大鼠急性肝损伤有一定的保护作用,其机制之一可能与提高机体抗氧化能力有关。Objective: To investigate the protective effects and the mechanisms of Jidesheng Sheyao (Sy), a Chinese patent medicine, on acute hepatic injury of rats. Methods: 48 SD rats were randomly divided into the normal control group, the model group, the high-dose treatment group of Sy and the low-dose treatment group of Sy. All experimental rats except those of the normal control group were given a single dose of Alpha-naphthylisothiocyanate(ANIT) to make a hepatic injury model, The serum biochemical indexes including alanine aminotransferase (ALT), aspartate aminotransferase (AST) and total bilirubin (TBil) of rats were monitored at 72h after the rats were given ANIT, and meanwhile, the contents of malondialdehyde(MDA), Glutathione-SH(GSH),nitric oxide (NO) as well as the activity of Superoxide dismutase(SOD) in serum were detected. The pathological changes of hepatic tissue of the rats were observed. Results: Compared with the model group, the serum levels of ALT of the rats of the low-dose treatment group significantly decreased and the serum levels of GSH obviously increased (P〈0.05). The hepatic pathological changes of rats of the treatment groups, especially of the low-dose group, were inferior to that of rats of the model group. Conclusion: Jidesheng Sheyao has some protective effects on ANIT-induced acute heDatic iniury of rats, one of the mechanisms of which may be related to enhancing antioxidation ability of the rat.
