IL-12、IL-7、IL-2协同诱导PBMC所产生的内源性细胞因子对其细胞毒活性的影响

摘　　要：应用细胞因子生物学检测法对ＩＬ－１２、ＩＬ－７、ＩＬ－２协同诱导的ＰＢＭＣ培养上清中的ＴＮＦ和ＩＬ－２进行了检测，同时观察了抗ＴＮＦ－α、抗ＴＮＦ－β和抗ＩＬ－２抗体对刺激细胞的细胞毒作用的影响。结果表明，ＩＬ－１２、ＩＬ－７、ＩＬ－２三种因子协同刺激ＰＢＭＣ可产生较高水平的ＴＮＦ，而抗ＴＮＦ－α单抗能显著抑制刺激细胞的杀瘤活性，提示内源性ＴＮＦ－α在介导该类细胞的细胞毒效应中起重要作用。The bioassay were used to estimate the level of TNF and IL2 in the supernatant of PBMC induced by IL12,IL7 and IL2.The effect of antiTNFα,antiTNFβ and antiIL2 antibody on cytotoxicity of PBMC were detected.The results showed that synergistic stimulation of IL12,IL7 and IL2 could produce high concentration of TNF in culture supernatant,and antiTNFα could obviously inhibit the cytotoxic activity of treated PBMC,therefore,the endogenous TNFα was considered playing important role in the cytotoxic activity of treated PBMC.
