T淋巴瘤侵袭转移诱导因子与大肠癌上皮间质转化的关系

摘　　要：目的探讨T淋巴瘤侵袭转移诱导因子1(Tiam1)与大肠癌上皮间质转化的关系。方法应用免疫组化二步法检测143例大肠正常组织、大肠癌、大肠淋巴结转移癌石蜡组织标本中Tiam1、E-cadherin、CK、vimentin的表达情况。结果(1)Tiam1在大肠癌组织中表达显著高于正常组织(P<0.01),且与大肠癌分化程度有关(P<0.05);Tiam1在淋巴结转移癌中表达显著高于大肠癌原发灶(P<0.05);(2)E-cadherin、CK在大肠正常组织中表达高于癌组织(P<0.01),在大肠癌中的表达与分化程度有关(P<0.01),但在大肠癌与淋巴结转移癌中表达差异无显著性;(3)vimentin在大肠癌中表达比正常组织高(P<0.01),且与分化程度有关(P<0.01),但在大肠癌与淋巴结转移癌中表达无显著性差异;(4)Tiam1与E-cadherin、CK表达呈负相关,与vimentin表达呈正相关。结论Tiam1与大肠癌分化和转移有关,其促进大肠癌转移的机制可能与上皮间质转化发生有关。Objective To investigate the relationship between T lymphoma invasion and metastasis 1 （Tiaml） and epithelial-mesenchymal transition （EMT） in human colorectal carcinomas. Method Tiaml, E-cadherin, CK, and vimentin expressions in normal colorectal epithelium, colorectal carcinomas （CRC） and CRC with lymphatic metastasis were determined by immunohistochemistry using a two-step method. Results Tiaml expression was significantly higher in CRC than in normal colorectal epithelium （P〈0.01） in close correlation to the degree of tumor differentiation （P〈0.05）. Higher Tiaml expression was detected in CRC with lymphatic metastasis than in primary CRC （P〈0.05）. The expressions of E-cadherin and CK in CRC tissues were significantly lowered in comparison with those in normal colorectal epithelium （P〈0.01）, showing a correlation to tumor differentiation （P〈0.01） but not to lymphatic metastasis. Vimentin was significantly overexpressed in CRC （P〈0.O1） and correlated to tumor differentiation （P〈0.01） but not to lymphatic metastasis. Tiaml expression was inversely correlated to E-cadherin and CK, but positively to vimentin. Conclusion Tiaml is related to the metastasis of colorectal carcinoma, and may induce EMT to promote CRC metastasis.
