摘 要:目的探讨术前介入化疗对胰腺癌部分肿瘤相关基因表达的影响,进一步了解胰腺癌介入化疗的分子机制。方法 8例行根治性切除术的胰腺癌标本,其中4例术前行介入化疗,另4例术前未行介入治疗作为对照,应用多点cDNA阵列分析法检测标本中9个肿瘤相关基因的表达差异,并通过RT-PCR法验证其中MBDI基因的表达变化,观察术前区域性动脉灌注化疗对胰腺癌部分肿瘤相关基因表达的影响。结果多点cDNA阵列分析结果显示:MBD1、Rb、E2F5、IGF1、TGF2B、CDH-1、RARB和RAB1术前介入组表达明显下降(P<0.05);RT-PCR进一步验证介入化疗使胰腺癌MBD1基因的表达水平明显降低(P<0.01)。结论术前介入化疗能够明显抑制胰腺癌中以MBD1基因为代表的多个肿瘤相关基因的表达,是胰腺癌综合治疗的有效措施,并为深入探讨介入化疗的分子机制提供了新思路。Object To investigate the effects of preoperative intra-arterial chemotherapy on the expression of tumor-related genes in pancreatic cancers, and to detect the molecular mechanisms of intra-arterial chemotherapy for pancreatic cancer. Methods The expression alterations of 9 tumor-related genes of 8 pancreatic cancer samples after radical resection were measured by means of multi-dots gene chip, among which 4 cases were treated by preoperative intra-arterial chemotherapy and the other 4 cases without preoperative treatment served as controls. The variation of expression of MBD1 was verified by method of RT-PCR, thus to further survey the effects of regional intra-arterial chemotherapy on relative genes expression in pancreatic cancer. Results It was demonstrated that the expression of genes such as MBD1, Rb, E2FS, IGF1,TGF2B, CDH-1,RARB and RAB1 were significantly decreased in patients treated with preoperative intra-arterial chemotherapy by muhi- dots gene chip analysis(P 〈 0.05 ). The MBD1 gene expression in pancreatic cancer was declined obviously after intra-arterial chemotherapy tested by RT-PCR( P 〈0.01 ). Conclusions Preoperative intra-arterial chemotherapy could suppress expression of tumor-related genes in pancreatic cancer. It might be an efficiency method of muhi-modality therapy for pancreatic cancer, and be helpful to understand the molecular mechanisms of regional intra-arterial chemotherapy.
