摘　　要：目的:观察Iressa对雌激素受体表达状态不同的人乳腺癌细胞:ER(+)的MCF-7和ER(-)的MDA-MB-231细胞增殖的抑制作用。方法:设置Iressa不同浓度组、时间组,采用四甲基偶氮唑蓝(MTT)法检测Iressa对细胞生长的影响,并分析量效关系、时效关系。结果:Iressa以0.01-1μmol/L浓度作用24h之后对2种细胞均有抑制增殖的作用,其中MDA-MB-231的抑制率于48h达到高峰;浓度为10μmol/L时抑制率高峰,MCF-7出现在48h,而MDA-MB-231在72h;且同浓度的Iressa作用相同时间后,对MDA-MB-231的抑制率更高。结论:一定浓度的Iressa作用一定时间后对雌激素受体表达状态不同的人乳腺癌细胞有抑制细胞增殖的作用,且对ER(-)的MDA-MB-231细胞作用更显著。Objective：To investigate the effect of Iressa on the proliferation in breast cancer cell lines：MCF-7 with ER（＋） and MDA-MB-231 with ER（-）.Methods：Establishing the breast cancer cell lines exposed to different concentration of Iressa for 24h,48h and 72h respectively,to observe the relation with time,dose and efficacy,the growth rate among these groups was detected by MTT assay.Results：Within the range of 0.01-1 μmol/L,Iressa couldn＇ t inhibit the proliferation of MCF-7 and MDA-MB-231 until after 24h.When cocultured with 10 μmol/L Iressa,the inhibitory ratio of MCF-7 and MDA-MB-231 achieved a peak after 48h and 72h respectively.Moreover,to compare with MCF-7 cells,MDA-MB-231 cells were more sensitive to Irresa under tne same concentration.Conclusion：Iressa could inhibit the proliferation of MCF-7 and MDA-MB-231 cells,the inhibition on MDA-MB-231 cells were more significant.
