摘 要:目的:研究胆囊癌及炎性组织细胞膜上生物大分子蛋白质及脂质的红外光谱改变。方法:应用傅里叶变换红外光谱技术(Fourier transformed infare spectroscopy,FTIR)对胆囊癌和炎性组织的细胞膜进行检测,并分析其蛋白质和脂质的红外变化。结果:胆囊癌和炎性组织细胞膜上生物大分子蛋白质和脂质的红外光谱存在明显差异:癌组织中酰胺Ⅱ带较弱、峰型低平,酰胺Ⅰ带较强、峰型宽大,相对吸收强度I1650/I1550明显升高;癌组织中与脂质相关的吸收谱带出现概率高、且峰型高尖,相对吸收强度I2957/I1650和I1740/I1400的比值增加;相对吸收强度I1460/I1400的比值在正常组织细胞膜中较高。结论:胆囊炎性与癌组织细胞膜上大分子红外光谱存在不同,通过FTIR能对细胞癌变早期生物大分子改变做出客观反映。Objective:To observe the changes of the protein and lipid in membrane of gallbladder cancer and cholecystitis by Fourier transform infrared spectroscopy(FTIR).Methods:The membrane of ten gallbladder cancer samples and ten cholecystitis tissue samples were observed by FTIR and data were summarized by Omnic 5.0 software.Results:Comparison with cholecystitis tissue,FTIR spectral of the cancerous menbrane were significantly difference.Alterations of bands related to protein:in the membrane of gallbladder cancer,the amideⅡ was weaker and lower,the amideⅠwas stronger and higher;The ratio of the I1650/I1550 raised.Alterations of bands related to lipid:in the membrane of gallbladder cancer,2957,2927,2853 and 1740 cm^-1 were stronger and higher,the ratio of the I2950/I1650 and I1740/I1400 raised,respectively.The ratio of theI1460/I1400 in the membrane of cholecystitis tissue was higher than the gallbladder cancer tissue.Conclusion:FTIR spectral of the membrane of cancer tissue are significantly different from those of cholecystitis,FTIR can early and objectively reflect the changes of biological macromolecules in the cancerous cells.
