摘  要:目的:研究黄荆子乙酸乙酯提取物(EVn-50)对人乳腺癌(T47D)细胞及其裸鼠移植瘤生长抑制的影响。方法:体外培养T47D细胞,溴化标记尿嘧啶(BrdU)掺入法测定细胞核酸合成的抑制作用;平皿克隆形成法测定细胞锚定依赖生长能力的抑制作用;人乳腺癌T47D裸鼠移植瘤模型治疗实验,观察EVn-50在体内对T47D细胞生长抑制作用,HE染色观察乳腺癌组织和细胞形态改变。结果:EVn-50抑制体外培养T47D细胞核酸合成和生长,呈剂量依赖性;EVn-50对裸鼠移植瘤的生长具有显著抑制作用,呈剂量和时间依赖性;EVn-5080mg/kg实验组对移植瘤的瘤重抑制率为51.4%(P<0.001);镜下观察EVn-5080mg/kg实验组肿瘤组织坏死组织较多,细胞异型性较少。结论:黄荆子乙酸乙酯提取物对人乳腺癌有生长抑制作用。Objective:To investigate the effect of the acetoacetate extract of Vitex Negu-ndo Seeds(EVn-50) on the growth inhibition of human breast cancer T47D cells and xe-nografts in nude mice.Methods:Human breast cancer T47D cells were cultured in vitro.The nucleic acid synthesis of EVn-50 on T47D cells was examined by BrdU incorporating assay.Clone-formation assay in plate was used to test inhibition of clone formation of T47D by EVn-50.Human breast cancer T47D xenografts in nude mouse model was esta-blished to investigate the growth inhibition of EVn-50.The morphology of tumor cells was observed by microscopy after HE staining.Results:EVn-50 inhibited nucleic acid synthesis and growth of T47D cells in a dose-dependent manner;EVn-50 inhibited the growth of human breast cancer T47D xenografts in nude mice,in a dose-and timedependent manner.The tumor weight inhibitory rate of EVn-50 80 mg/kg was 51.4%.The morphologic observation exhibited larger areas of necrosis and less degree of atypia after EVn-50 treatment.Conclusion:EVn-50 inhibited the growth of human breast cancer in vitro and in vivo.
