摘 要:目的构建适合移植使用的BALB/c-nu/nu裸鼠造血微环境损伤模型。方法分别用3.0、4.0、5.0、6.0、7.0Gv剂量辐照BALB/c-nu/nu裸鼠,检测照射后1、3、5、7、14、21d血象变化,检测照射后1、7、14、21d基质细胞集落形成单位(CFU.F)和巨核细胞计数,观察照射后骨髓组织活检变化情况。结果7.0Gy辐照剂量照射后裸鼠全部死亡;6.0Gy组照射后出现明显骨髓抑制,至照射后21d不能恢复至照射前水平,且部分裸鼠照射后死亡;3.0、4.0Gy照射后裸鼠仅出现轻度骨髓抑制;5.0Cy组照射后出现明显骨髓抑制,未做特殊处理于照射后21d自行恢复至照射前水平。不同剂量放射线照射后裸鼠CFU-F计数变化不同。3.0Gy和4.0Cy组CFU-F在照射后7~14d出现轻度减少,6.0Gy组照射后CFU-F明显减少,至照射后21d不能恢复,5.0Cy组照射后CFU-F也出现了较明显减少,但在照射后21d恢复至照射前水平。结论5.0Gy是BALB/c-nu/nu裸鼠造血微环境损伤模型的适宜照射剂量。Objective To establish hematopoietie microenviroment damage model in BALB/e-nu/nu nude mice for transplant experiment. Methods Experimental nude mice were irradiated with 3.0, 4.0, 5.0, 6.0 and 7.0 Gy 60Co γ ray respectively. Then the hemogram changes were detected at 1, 3, 5, 7, 14, 21 d after irradiation, the quantity of CFU-F and megakaryocyte was counted at 1, 7, 14, 21 d after irradiation, and the biopsy of myeloid tissue was performed. Results Irradiated with 7.0 Gy, all nude mice died. Bone marrow of nude mice irradiated with 6.0 Gy was depressed obviously, and did not recover within 21 d, and some of them died after irradiation. Slight bone marrow depression was observed in the nude mice irradiated with 3.0 Gy and 4.0 Gy. Nude mice irradiated with 5.0 Gy showed obvious bone marrow depression, but it recoverd within 21 d. CFU-F count had slight decrease in the nude mice irradiated with 3.0 or 4.0 Gy, and remarkable decrease in the nude mice irradiated with 6.0 Gy till the 21 st day after irradiation. CFU-F count also had remarkable decrease after irradiation with 5.0 Gy and recovered in 21 d after irradiation. Conclusion The suitable radiation dose is 5.0 Gy for creating hematopoietic microenviroment damage model in BALB/c-nu/nu nude mice.
