摘  要:目的探讨抗抑郁药物对抑郁模型大鼠海马血管内皮细胞生长因子(vascular endothelial cell growth factor,VEGF)的作用及机制。方法选择成年雄性SD大鼠40只,分为应激+氟西汀组(氟西汀组)、应激+噻奈普汀组(噻奈普汀组)、应激组(模型组)、应激+生理盐水组(盐水对照组)、空白对照组,每组8只。采用孤养与慢性不可预见性中等强度应激(chronic unpredieted mild suress,CUMS)联合建立抑郁模型,旷场分析和液体消耗实验进行行为学观察。对抑郁模型进行抗抑郁药物氟西汀和噻奈普汀以及生理盐水作为对照灌胃30d,免疫组化观察各组大鼠海马VEGF表达情况。结果氟西汀组、噻奈普汀组与生理盐水对照组通过较旷场分析、液体消耗试验显示抑郁模型成功,免疫组化图像分析氟西汀组、噻奈普汀组与生理盐水对照组大鼠海马VEGF表达有统计学意义(P<0.05)。结论抗抑郁药物氟西汀、噻奈普汀均能促进抑郁模型大鼠海马VEGF的表达,VEGF可能参与抑郁症的病理生理机制。Objective To study the mechanism by which antidepressant works on vascular endothelial cell growth factor (VEGF) in rat hippocampus of depression model. Methods Forty SD rats were randomly and equally divided into 5 groups: stress + fluoxetine, stress + tianeptine, stress, stress + normal sodium, blank matched control. We adopted separation and chronic unpredicted mild stress to establish depression in all rats except those in blank matched control group, and the behavior of all animals was evaluated by open-field and fluid consumption test. VEGF expression was detected by immunohistochemisty after intragastric administration respectively with fluoxetine, tianeptine or normal sodium for 30 d. Results VEGF expression in hippocampus was decreased in all stress groups compared with control group (P 〈 0.05 ). After antidepressant administration for 30 d, VEGF expression in hippocampus was increased in fluoxetine group and tianeptine group compared with normal sodium group (P 〈 0.05). Conclusion Chronic antidepressant treatment significantly increases VEGF expression in hippocampus of rat depression model, suggesting VEGF may be correlated with the pathophysiology of depression.
