摘  要:目的观察大鼠脑缺血再灌注后缺血半暗带皮质Fas相关的磷酸酯酶-1(FAP-1)mRNA及蛋白的表达变化。方法用半定量的逆转录PCR(RT-PCR)法检测缺血2h再灌注不同时间点缺血半暗带皮质内FAP-1mRNA的表达,免疫组化法检测FAP-1蛋白表达的变化。结果缺血半暗带脑皮质FAP-1 mRNA及其蛋白的表达于缺血灌注后再灌注1h开始明显上升,6h达高峰,24h显著下降。结论脑缺血再灌注后缺血半暗带皮质内FAP-1 mRNA及蛋白表达均明显增加,提示其可能参与了脑缺血后抗凋亡作用。Objective To study the mRNA and protein levels of Fas associated phosphatase-1 ( FAP-1 ) in the penumbra of rat cortex after transient focal cerebral ischemia. Methods One hundred male Sprague-Dawley rats were subjected to the left middle cerebral artery occlusion for two hours, then killed at 1,3,6,12 and 24h after reperfusion. The mRNA level of FAP-1 was observed by half-quantitative RT-PCR. The protein level of FAP-1 was measured with meathod of immunohistochemistry. Results The mRNA and protein levels of FAP-1 in the penumbra of rat cortex were all increased at lh after ischemia,and peaked at 6h after reperfusion (P 〈 0. 01 ), and then markedly declined at 24h after reperfusion. Conclusion The mRNA and protein levels of FAP-1 increase in the penumbra of rat cortex after transient focal cerebral ischemia which suggests that FAP-1 may inhibit neuronal apoptosis following cerebral ischemia.
