摘 要:目的观察肝硬化大鼠AVP系统的变化及中药黄芪的治疗作用。方法采用斑点杂交和半定量RT-PCR法检测胆总管结扎2周所致肝硬化大鼠下丘脑AVP及其肾脏皮质V1a受体基因表达。结果肝硬化大鼠下丘脑AVPmRNA水平明显升高,肾脏皮质V1a受体mRNA表达下调,可能与非渗透性因素有关。应用黄芪可使模型鼠AVP高表达下降,并促使肾脏皮质V1a受体基因表达进一步降低。结论肝硬化大鼠存在AVP系统异常,黄芪具有一定改善作用。Objective To investigate the changes of AVP system in liver cir- rhosis and the role of astragalus membranaceus(AM). Method Dot blot analysis and relatively quantitative RT-PCR technique were used to determine the levels of AVP mRNA in hypothalamus and V1a receptor mRNA in renal cortex of cirrhotic rats in- duced by common bile duct ligation for 2 weeks. Results The cirrhotic rats demon- strated obviously enhanced expression of AVP mRNA in hypothalamus and weakened AVP V1a receptor mRNA expression in renal cortex. These changes may be through non-osmotic mechanisms.AM could alleviate the over expression of hypothalamic AVP V1a receptor mRNA expression in renal cortex.These changes may be through rhotic rats. Conclusioa The abnormality of AVP system exists in liver cirrhosis ani- mals, AM has a therapeutic effect.
