摘  要:将构建的编码乙肝病毒PreS2+S蛋白的真核表达质粒注射于C57BL/6小鼠胫前肌内,3d后,在同一部位注射rhGM-CSF。基因疫苗接种2周后,血清中可测到抗-HBs,两月后,抗体水平达到高峰,并保持高水平至少2月以上;肌内注射GM-CSF可提高血清抗-HBs水平,提示GM-CSF可作为基因疫苗的佐剂。The eukaryotic expression plasmid encoding HBV envelop PreS2+S protein was injected into tibialis anterior muscles of C57BL/6 mice. After 3 days of genetic vaccine inoculation, re-combinant rhGM-CSF was administered at the same sites. ELISA was used to detect the anti-HBs in mice sera at various times after gene transfer. It was found that anti-HBs was detected in sera of mice (≥10mIU/ml) 2 weeks after plasmids injection. The level of antibody attained the peak two months after genetic vaccination and kept in high titer for at least 2 months ; rhGM-CSF injected intramuscularly could significantly (P<0. 01) promote the production of anti-HBs induced by HBV DNA vaccine. The results suggested that GM-CSF can act as an immunological adjuvant for genetic vaccines.
