摘  要:目的:探讨β2微球蛋白(β2-MG)对非霍奇金淋巴瘤(NHL)患者树突状细胞(DC)分化成熟和功能的抑制作用。方法:采集NHL患者骨髓单个核细胞,经GM-CSF、IL-4和TNF-γ诱导DC生成。于DC培养第1天起加入β2-MG设为实验组A,DC培养第4天起加入β2-MG设为实验组B,将同时段DC加入人血白蛋白分别设为对照组A和B,设空白对照组。光镜下观察DC形态;流式细胞术检测DC表型;培养DC与自身淋巴细胞共孵育,ELISA法检测IFN-γ和IL-10水平。结果:与其他4组相比,实验组A的DC体积小,无明显树突状突起。表型分析显示,除CD1a外,CD83、CD80、CD86和HLA-DR的表达均明显降低(P<0.05);激活T细胞分泌IFN-γ水平下降(P<0.05),分泌IL-10的水平升高。结论:β2-MG可以抑制NHL患者DC的分化成熟及T细胞活化,可能对揭示NHL的进展机制及进一步免疫治疗具有重要意义。Objective:To investigate the immune reactive of dendritic cells (DCs) inhibited by β2-microglobulin(β2-MG) for non-Hodgkin lymphomas. Methods:DCs were cultured from bone marrow mononuclear cells of the new-diagnosed non-Hodgkin lymphomas patients by the conventional methods. On the first day, β2-MG-treated DC, human serum albumin-pulsed DC and unpulsed DC served as experimental groups, control groups and blank group, respectively. On the forth day, experimental group B, countrol group B and blank group were established in the same way. DCs morphous was observed per every other day under light microscope. The immune phenotypes of DCs were analysed by flow cytometry. After DCs incubated with auto-T lymphocytes, the levels of IFN-3, and IL-10 in cultural supernatants were detected by ELISA. Results:Under light microscope, the cells concuhured with β2-MG did not have the feature of DC-like morphology on day 8. The levels of mature phenotypes on DCs (CD80, CD83, CD86 and HLA-DR) cultured with β2-MG were down-regulated. Also, the level of IFN-γof experimental group A was lower than other groups while the level of IL-10 on the contrary(P 〈 0. 05). Conclusion: β2-MG could inhibit DCs maturing and auto-T lymphocytes to be activated.
