摘  要:目的:研究肥大细胞在实验性哮喘小鼠气道病理生理改变过程中的作用。方法:遗传性肥大细胞缺失W/Wv小鼠、正常同源基因+/+小鼠和培养骨髓肥大细胞重建的W/Wv小鼠分设对照组和实验组;实验组小鼠用10μgOVA两次致敏,17d后用1%OVA气道激发,隔天1次,共3次;第22天时检测其气道反应性,后取材观察其肺组织中肥大细胞数量、和肺组织的病理改变。结果:OVA致敏和激发可引起+/+小鼠和肥大细胞重建的W/Wv小鼠气道反应性增强,肺组织中肥大细胞数量明显增加,肺组织炎性病理改变明显;而OVA致敏和激发的W/Wv小鼠的上述指标改变不明显。结论:肥大细胞参与了OVA诱导的小鼠气道高反应性、肺组织炎症细胞浸润过程。Objective : To investigate the role of mast cells in the physiopathology in a murine asthma model. Methods: Genetical mast cell deficient W/Wv mice,their congenic normal littermates +/+ mice and BMCMC→W/Wv mice were randomly divided into control and experimental groups;experimental mice were sensitized with 10μg OVA , 2 times; 17 days after the beginning of the sensitization the mice received their airway challenges with 1% OVA, 3 times;later,their airway reactivity, lung mast cell infiltration,and lung pathologic change were studied. Results : OVA sensitization and airway challenge could remarkably amplify the airway reactivity, increase the number of lung mast cells and the lung pathologic inflammatory change in the +/ + and BMCMC→W/Wv mice; but the changes in OVA sensitized and challenged W/Wv mice were not obvious. Conclusion: Mast cells take part in the airway high reactivity and lung inflammation in the experimental OVA-induced murine asthma.
