摘 要:目的:探讨肺气肿发生过程中胆红素对纤维黏连蛋白(Fn)的影响。方法:将健康Wistar大鼠随机分为模型、干预、对照3组。模型组:被动吸烟6月。干预组:吸烟同模型组,熏烟前予间接胆红素灌胃。6月末处死动物,取材观察组织学结构;支气管肺泡灌洗液(BALF)及血清中纤维黏连蛋白(Fn)、丙二醛(MDA)与谷光甘肽(GSH)含量;间接免疫荧光法测定肺泡隔Fn表达。结果:组织学结构观察:模型组符合肺气肿病理表现,模型复制成功。干预组大鼠的肺泡隔断裂和肺泡腔扩大情况较模型组减轻;模型组BALF及血清中Fn、MDA含量明显高于对照组(P均<0.05);干预组BALF中Fn、MDA含量低于模型组(P均<0.05);模型组BALF及血清中GSH含量明显低于对照组(P均<0.05);干预组BALF中GSH含量高于模型组(P<0.05);相关分析显示BALF中的MDA与GSH含量呈负相关;肺泡隔Fn免疫荧光表达在模型组强于对照组(P<0.05),干预组弱于模型组(P<0.05)。结论:肺气肿形成过程中伴随着错综复杂的Fn的变化,胆红素干预可使其变化趋于正常;胆红素具有抗氧化作用,对机体氧化抗氧化平衡的维持有重要作用。Objective: To investigate the influence of bilirubin on expression of Fn in rat pulmonary emphysema model. Methods:Wister rats were randomly divided into three groups, the control group , the pulmonary emphysema model group (the model group) and the bilirubin treated group (the treated group). The model group was established by chronic exposure to cigarette smoke. The treated group was treated by bilirubin through feeding before the pulmonary emphysema model had been established. All of the rats were sacrificed to be tested at the end of six months. The histological changes were evaluated. The levels of Fibronectin(Fn), Malonyl dialdegyde (MDA) and Glutathione peroxidase (GSH) of broncho alveolar lavage fluid (BALF) and serum were determined. The expression of Fn was measured by immunofluorescence method. Results:From histological observation, the pathological change in model group was accorded with those of pulmonary emphysema,and the change in the treated group was slight obviously. The Fn and MDA levels of BALF and serum in model group was significantly higher than those in control group(P〈0.05). Those of BALF in treated group were significantly lower than those in model group(P〈0.05). Significant decrease of the level of GSH in BALF and serum were found in model group in comparison with that in the control group (P〈 0. 05), and significant increase of the level of GSH in BALF was found in treated group as compared with that in the model group(P〈0.05). Significant negative correlation was observed between the levels of MDA and GSH of BALF (r=-0.81, P〈0.05). The expression of Fn in the alveolar was significantly increased in model group than in control group (P〈 0. 05)and those in treated group was significantly decreased than those in control group. Conclusion:There are complex changes of Fn during the form of pulmonary emphysema in rats ,but bilirubin can prevent the development of pulmonary emphysema. Antioxidation can be provided by biliru
