左旋肉碱对顺铂损伤大鼠肾超微结构的影响

摘　　要：目的:观察左旋肉碱(LC)对顺铂损伤大鼠肾病理和超微结构的影响。方法:将36只成年雌性W istar大鼠随机分为正常对照组(Control组),化疗模型组(DDP组),干预组(LC+DDP组)。Control组和DDP组给予常规饲料,LC+DDP组给予常规饲料+1%LC。喂饲7 d后,DDP组和LC+DDP组腹腔注射顺铂(5 mg/kg),Control组腹腔注射生理盐水。注射4 d后,取标本,光镜和透射电镜下观察大鼠肾脏形态结构改变。结果:病理切片显示,DDP组肾小球结构尚完整,近曲小管上皮细胞有变性、坏死,肾小管出现蛋白管型;而LC+DDP组肾脏病理结构明显改善。超微结构观察,DDP组近曲小管上皮细胞线粒体高度肿胀,嵴断裂缺失,部分呈空泡变性,部分崩解;而LC+DDP组这些损害轻微。结论:预防性喂饲LC能对顺铂损伤大鼠肾病理和超微结构产生一定的保护作用。Objective： To observe the effects of L - carnitine （ LC ） on the renal uhrastructure damage induced by cisplatin （DDP）. Methods ： Female adult Wistar rats were randomly divided into control group, DDP group, LC ＋ DDP group and each group consisted of 12 animals. Rats in control group and DDP group were fed a common diet while those in LC ＋ DDP group were added 1% LC into their common diet. After feed for 7 days, animals in DDP group and LC ＋ DDP group received cisplatin intraperitoneal injection at 5 mg/kg body weight while those in control group were given normal saline. On the fourth day after injection, all rats were killed, and the renal morphological structures were observed under light microscope and transmission electron microscope （TEM）. Results：The pathological slices indicated that the glomcrulus structures were complete in DDP group,but the vacuolar degeneration and necrosis in endothelial cells of proximal convoluted tubule and protein casts in renal tubules were found. However, these changes were significantly improved in LC ＋ DDP group. Furthermore, uhrastructures under TEM demonstrated that swelling,vacuolar degeneration, disaggregation and cristae fragmentation and absence of mitochondria were less apparent in LC ＋ DDP group than those in DDP group. Conclusion： To some extent, a preventative dietary supplementation with LC might protect renal pathological structure and uhrastructure from the damages induced by cisplatin in rats.
