激素性高眼压患者外周血单个核细胞糖皮质激素受体α、β的表达及意义

摘　　要：目的检测糖皮质激素性高眼压患者外周血单个核细胞(PBMCs)内糖皮质激素受体(GR)亚型GRα和GRβmRNA的表达,并探讨其与激素性青光眼的关系。方法收集9例激素性高眼压患者(高眼压组),10例用激素后眼压正常患者(正常眼压组),10例健康志愿者(对照组)的外周血,提取单个核细胞RNA,半定量逆转录-聚合酶链反应(RT-PCR)法检测PBMCs内GRα、GRβmRNA表达水平。结果外周血单个核细胞有GRα、GRβmRNA表达,以GRα为主。高眼压组的GRαmRNA半定量比值为(1.11±0.17),与对照组(1.07±0.09)和正常眼压组(1.06±0.11)相比,其表达增高,差异有显著性意义(均P<0.05)。高眼压组、正常眼压组、对照组的GRβmRNA半定量比值分别为(1.030±0.004)、(1.040±0.004)、(1.030±0.006),3组之间的表达差异无显著性意义(均P>0.05)。结论外周血GRαmRNA的表达水平与糖皮质激素性青光眼的发病密切相关,外周血GRβmRNA与激素性青光眼的关系可能不大,但GRβ的作用仍需从其他方面继续研究。Objective To detect the expression of α- and β-isoforms of glucocorticoid receptors （GR）, and to explore their relationship with steroid-induced glaucoma. Methods Peripheral blood from 9 patients with glaucocorticoid ocular hypertension （ocular hypertension group）, 10 patients without ocular hypertension after steroid treatment （normal ocular pressure group） and 10 healthy volunteers （control group） was collected to detect the expression level of GRα and GRβ mRNA in peripheral blood mononuclear cells （PBMCs） by semi-quantitative reverse transcriptional polymerase chain reaction （RT-PCR）. Results There were both GRa and GRβ mRNA in PBMCs, and GRα was predominant. GRα/GAPDH in ocular hypertension group was （1.11~0.17）, which was significantly higher than that of normal ocular pressure group （1.06 ~ 0. 11） and control group （1.07___0.09） （P〈0.05）. The expression of GRα mRNA was detected in ocular hypertension group, normal ocular pressure group and control group with GRβ/GAPDH being （1. 030±0. 004）, （1. 040±0. 004）, （1. 030±0. 006） respectively, and no significance difference was found among the three groups. Conclusion GRα expression in PBMCs may have a close relationship with steroid-induced glaucoma.
